Abstract

Bee pollen has been reported to show antioxidant and radical scavenging activities; contributing to anti-inflammatory and gastroprotective properties. Venezuelan honeybee pollen has been little studied, but is consumed because its properties are known from other countries reports. On the basis of these reports, water, ethanol and methanol soluble fractions were prepared from dried bee-pollen commercially available and produced by La Montaña farm (Mérida, Venezuela). These fractions were evaluated for their functional properties, specifically, polyphenol content and total antioxidant activity. Pollen samples were separated by color in four fractions: yellow, brown, orange and ochre. Polyphenol content ranged between 396.7 to 1286.7 gallic acid equivalents GAE/100 g pollen; it was highest in pollen homogenates obtained with ethanol, followed by those obtained with methanol and water. The antioxidant activity ranged from 0.50 to 1.84 micromoles Trolox equivalents TEAC/100 g for water and ethanol homogenates respectively. The results presented in this work suggest that the ethanol extract of bee pollen show a potent antioxidant activity, comparable to human plasma, probably due to total polyphenol content of bee pollen. This is important because the bee pollen would be beneficial not only as a dietary supplement but also as a functional food.

Abstract

BACKGROUND:

Bee pollen, a honeybee product, is the feed for honeybees prepared themselves by pollens collecting from plants and has been consumed as a perfect food in Europe, because it is nutritionally well balanced. In this study, we aimed to investigate the anti-inflammatory effect of bee pollen from Cistus sp. of Spanish origin by a method of carrageenan-induced paw edema in rats, and to investigate the mechanism of anti-inflammatory action and also to elucidate components involved in bee pollen extracted with ethanol.

METHODS:

The bee pollen bulk, its water extract and its ethanol extract were administered orally to rats. One hour later, paw edema was produced by injecting of 1% solution of carrageenan, and paw volume was measured before and after carrageenan injection up to 5 h. The ethanol extract and water extract were measured COX-1 and COX-2 inhibitory activities using COX inhibitor screening assay kit, and were compared for the inhibition of NO production in LPS-stimulated RAW 264.7 cells. The constituents of bee pollen were purified from the ethanol extract subjected to silica gel or LH-20 column chromatography. Each column chromatography fractions were further purified by repeated ODS or silica gel column chromatography.

RESULTS:

The bee pollen bulk mildly suppressed the carrageenan-induced paw edema and the water extract showed almost no inhibitory activity, but the ethanol extract showed relatively strong inhibition of paw edema. The ethanol extract inhibited the NO production and COX-2 but not COX-1 activity, but the water extract did not affect the NO production or COX activities. Flavonoids were isolated and purified from the ethanol extract of bee pollen, and identified at least five flavonoids and their glycosides.

CONCLUSIONS:

It is suggested that the ethanol extract of bee pollen show a potent anti-inflammatory activity and its effect acts via the inhibition of NO production, besides the inhibitory activity of COX-2. Some flavonoids included in bee pollen may partly participate in some of the anti-inflammatory action. The bee pollen would be beneficial not only as a dietary supplement but also as a functional food.

Abstract

The effect of bee pollen on productive and reproductive performances of adult buck rabbits and their offspring was studied during winter and summer seasons. Forty New Zealand White bucks were equally divided among four groups feeding the same commercial diet and receiving a water solution containing, respectively, 0 (control), 100, 200 and 300 mg bee pollen/kg body weight, twice per week along two experimental periods. The experimental periods were listed for ten weeks both during winter (30-40 weeks of age) and summer seasons (56-66 weeks of age). During the trials body weight, body weight gain, total feed intake, semen quality, fertility and blood constituents were determined. Fertility was determined after natural mating with no treated females. For each season, 80 weaned rabbits obtained from the bucks of the control group were equally divided (20 per group) among 4 levels (0, 100, 200 and 300 mg/kg BW) of bee pollen, given as a water solution twice per week. The offspring sired by bucks given 100, 200 and 300 mg (20 for each group and season) were not administrated bee pollen. The effect of bee pollen on growth performance of rabbits was studied from 4 to 12 weeks of age. Bee pollen at 200 mg/kg BW significantly (p< 0.01) improved semen quality, increased fertility percentage, improved biochemical profiles of blood and helps outstanding of bucks during both seasons. The same concentration of bee pollen increased body weight gain and survival rate and reduced feed intake and feed conversion ratio of offspring up to 12 weeks of age.

Abstract

Bee pollen of Brassica campestris L. is widely used in China as a natural food supplement and an herbal medicine in strengthening the body's resistance against diseases including cancer. The present study was carried out to investigate the effect of a steroid fraction of chloroform extract from bee pollen of Brassica campestris L. on human cancer cell viability. Our studies show that among nine cancer cell lines of different origin (PC-3, LNCaP, MCF-7, Hela, BEL-7402, BCG-823, KB, A549 and HO8910), this steroid fraction displayed the strongest cytotoxicity in human prostate cancer PC-3 cells. The mode of cell death appeared to be apoptosis in PC-3 cells, as shown by flow-cytometric analysis and fluorescence microscopes. Caspase-3 activity was obviously enhanced after the cells were treated with the fraction. A time-dependent decrease in the expression of anti-apoptotic protein Bcl-2 was also observed by Western blot analysis. It is suggested that the steroid fraction could induce cytotoxicity in prostate cancer PC-3 cells by triggering apoptosis. The studies indicate that the steroid fraction of chloroform extract from bee pollen of Brassica campestris L. may be a promising candidate for the treatment of advanced prostate cancer.

Abstract

Bee-collected pollen and propolis are apicultural products which are composed of nutritionally valuable substances and contain considerable amounts of polyphenol substances which may act as potent antioxidants. We wanted to show if respiratory burst within a cancer cell lines could be influenced when incubated with pollen and propolis extracts or not. Pollen and propolis extracts at concentrations of 50, 25, 12.5 and 0 mg/ml were prepared by dimethyl sulfoxide (DMSO). K-562 cell cultures and mononuclear cell (MNC) cultures prepared from a peripheral blood sample to serve as control cells were incubated with extracts for 24 h. Determination of respiratory burst was carried out by intracellular dichlorofluorescein (DCFH) test by using flow-cytometric fluorescence analysis. While about 90% and 66% fluorescence was detected at zero concentrations for both K-562 and MNC cultures, fluorescence positivity decreased (between 3.8% and 11.8%) as concentrations of both propolis and pollen extracts increased for K-562 cell culture, but unchanged (between 20% and 83%) for MNC culture. It was concluded that pollen and propolis extracts inhibit respiratory burst within cancer cell lines probably by their antioxidant potentials.

Abstract

Bee pollen is considered, since memorable times, a good source of nourishing substances and energy. The present study aimed to evaluate the biological activities of eight commercial bee pollens purchased from the market. The origin of sample A was not specified in the labeling; samples B, C, D and G were from Portugal and the remaining were from Spain. The sample E presented the highest value of phenolics (32.15±2.12 mg/g) and the H the lowest (18.55±095 mg/g). Sample C had the highest value of flavonoids (10.14±1.57 mg/g) and sample H the lowest (3.92±0.68 mg/g). All the samples exhibited antimicrobial activity, being Staphylococcus aureus the most sensitive and Candida glabrata the most resistant of the microorganisms studied. All the samples exhibited antimutagenic activity, even though some samples were more effective in decreasing the number of gene conversion colonies and mutant colonies. Regarding the antioxidant activity, assessed using two methods, the more effective was sample B. The anti-inflammatory activity, assessed using the hyaluronidase enzyme, was highest in samples B and D. Pearson's correlation coefficients between polyphenols, flavonoids, antioxidant activity and antimicrobial activity were computed. It was also performed a discriminant analysis.
